He was a retainer and close advisor of John Mowbray, Duke of Norfolk. He was knighted in 1471 at the battle of Tewkesbury. He was present at the 1483 coronation of Richard III, but his sons William and Thomas joined Buckingham's rebellion in the same year, and some of his lands were seized by the Crown. He secured a pardon in 1484, but subsequently found himself out of favor; he took sanctuary in Gloucester until the battle of Bosworth.
He may have been a son of Robert Brandon (d. ~1438), collector of customs at King's Lynn and Great Yarmouth, Norfolk, son of John Brandon, merchant of and collector of customs at King's Lynn.
William Brandon and Elizabeth Wingfield were great-grandparents of Lady Jane Grey:
